Anti-Inflammatory Effect of Chondria crassicaulis Ethanol Extract on MAPKs and NF-¥êB Signaling Pathway in LPS-Induced RAW 264.7 Macrophages

Abstract
The anti-inflammatory effect of Chondria crassicaulis ethanol extract (CCEE) was investigated using croton oil-induced ICR mice and LPS-induced RAW 264.7 cells. The CCEE showed inhibitory effect on the production of proinflammatory cytokines (IL-6, IL-1¥â, and TNF-¥á) and nitric oxide (NO) in LPS-induced inflammatory response as well as inducible nitric oxide, cyclooxygenase-2 via inactivation of mitogen-activated protein kinases (MAPKs) and nuclear factor- kappa B (NF-¥êB). In croton oil-induced ear edema test, the edema formation of CCEE treatment group (10-250 mg/kg body weight) was lower than that of the positive control and 250 mg/kg body weight treatment group appeared similar effects compared with prednisolone (10 mg/kg body weight). The results in photomicrograph of mice ear tissue showed the reduction in dermis thickness and the number of infiltrated mast cells. Therefore, these results indicate that CCEE exerts anti-inflammatory effect via inhibition of NF-¥êB and MAPKs activation and can be applied as a natural anti-inflammatory resource.
